I just had the same problem, and I got to fiddling around with the files and fixed it. You need to go into Documents > My Games > Fable > Saves > (your profile name), and delete everything but profile.bin and your latest manual save, it should look like this: "Manual-Save1. Then start up the game and it should work, though it still loads slow.
